Biosafety Cabinet Classes Compared for Federal Labs

A biosafety cabinet class states who gets protected. Class I protects the operator and the room, not the sample. Class II protects operator, room and sample. Most solicitations ask for "a Class II biosafety cabinet" and stop. That sentence buys four different machines, and the difference is measured in ductwork. Class Answers One Question: Who Is Being Protected?
Class I is the simplest of the three. Room air pulls in across the front opening, passes over the work, and leaves through a high-efficiency particulate air (HEPA) filter. Operator and environment are protected. The sample sits in unfiltered room air. Good enclosure for a centrifuge. Wrong device for cell culture.
Class II adds a HEPA-filtered downflow over the work surface, and that downflow protects the product. Inward air at the front still protects the operator, exhaust is still HEPA-filtered. When a biomedical lab says "biosafety cabinet," it means Class II.
Class III is a sealed, gas-tight enclosure. The operator works through attached gloves, supply air is HEPA-filtered, and exhaust passes through two HEPA filters in series or a filter plus thermal treatment. If a spec calls for Class III and the room was not designed around it, the spec is ahead of its building.
Class II Types Are Where a Specification Breaks
Each line below gives the Class II type first, then its exhaust arrangement, then what that arrangement costs the buyer.
Type A2: exhausts through a HEPA filter back into the room, or through a canopy connection to building exhaust. It holds its own airflow whether or not the building exhaust runs, which is why most labs can install one without mechanical construction.
Type B1: hard-ducted to a dedicated exhaust, recirculating part of the downflow and exhausting air drawn across the rear of the work surface. Only that directly exhausted portion can take volatile chemistry.
Type B2: total exhaust, no air recirculated inside the cabinet or back into the room. Largest mechanical scope, largest conditioned-air penalty, and the type for volatile chemicals or radionuclides.
Type C1: convertible, running recirculating or connected to exhaust as the work changes. It answers a room whose protocols aren't settled, and costs more than an A2 to buy and commission.
The uncomfortable reading: an A2 on a canopy connection covers most federal biomedical work, and B2 gets specified more often than the chemistry justifies. A B2 pulls conditioned air out of the room continuously, and the heating, ventilating and air conditioning (HVAC) system replaces every cubic foot. That cost lands on somebody who never sees the utility bill.
Exhaust Configuration Decides How Much Building You Buy
A canopy connection, sometimes called a thimble, sits over the cabinet's exhaust with a deliberate air gap. Building exhaust pulls from the room through that gap, so a fluctuation upstream doesn't upset airflow inside the cabinet. Balancing is simpler, and a stumble in the exhaust fan doesn't break containment at the work surface.
A hard duct is a different relationship. Cabinet and building exhaust become one system. That buys a B1 or a B2, and brings interlocks, alarms, commissioning coordination, and a rebalance whenever somebody adds a device to that branch. The same logic governs chemical hoods, worked through in ducted versus ductless fume hoods. The person writing the cabinet line rarely owns the exhaust riser, so get both in one room before the type is fixed.
Volatile Chemicals Are a Separate Permission
A biosafety cabinet is a biological containment device. Its HEPA filters capture particles, not vapor. A cabinet recirculating into the room returns any solvent vapor put inside it, which is why an A2 in recirculating mode is not where volatile chemistry belongs.
Where volatile work is permitted at all, it belongs in the directly exhausted portion of a B1, throughout a B2, or in an A2 or C1 connected to exhaust, and only in quantities the manufacturer and biosafety officer agree to. Appendix A of the Centers for Disease Control and Prevention and National Institutes of Health publication Biosafety in Microbiological and Biomedical Laboratories (BMBL), 6th Edition covers selection, installation and use of these cabinets. BMBL is advisory. It binds when an agency adopts it or a contracting officer writes it into the solicitation.
Chemical exposure stays governed on its own track. The Occupational Safety and Health Administration (OSHA) standard at 29 CFR 1910.1450 requires a written Chemical Hygiene Plan and requires that protective equipment function. A biosafety cabinet doesn't discharge that obligation, and was never built to.
Field Certification Is an Award Line Item
Every Class II cabinet gets certified on site to NSF/ANSI 49, the voluntary consensus standard for biosafety cabinetry, by an accredited field certifier after final placement. Again after any relocation, including a move across the same room. Again after a filter change. A factory certificate is not a substitute.
It is a scheduled service with a technician, an appointment and a written report. Missing from the award, it returns as a change order while the room sits unusable. Put certification on the cabinet's own line, and name who holds the recurring cycle.

What is the difference between a Class I and a Class II biosafety cabinet?
Both protect operator and environment with inward airflow and a HEPA-filtered exhaust. Only Class II protects the sample, because only Class II supplies HEPA-filtered downflow over the work surface. Class I bathes the work in room air, so anything sterile rules it out.

Not in the way a B1 or B2 is ducted. A canopy leaves an intentional air gap between cabinet exhaust and building duct, so the cabinet keeps its own airflow. A hard duct ties cabinet performance to the building exhaust and brings interlocks, alarms and coordinated commissioning.
